Cytotoxic effects on cancerous and non-cancerous cells of trans-cinnamaldehyde, carvacrol, and eugenol

Abstract Essential oils and their active components, referred here as plant derived antimicrobials (PDAs), have been used for antimicrobial, anti-inflammatory antioxidant properties. Many reports also document PDAs’ cytotoxic effects on cancerous cells, raising the hope that they could be cancer treatments. Due to lack of specificity, we hypothesize PDAs are both non-cancerous cells. Trans -cinnamaldehyde (TCA), carvacrol, eugenol were assessed cytotoxicity HeLa cells normal skin fibroblasts (CCD-1123Sk, CCD) by MTT LDH assays, flow cytometry, reverse transcription quantitative PCR (RT-qPCR). After 24 h treatment, carvacrol TCA significantly decreased cell viability (by more than 50%) at 100 µg/ml, whereas was ineffective up 400 µg/ml. Cell detachment increased apoptosis observed with µg/ml types. RT-qPCR apoptotic genes ( BCL2, CASP3 CASP8 ) necrosis MLKL, RIPK1 RIPK3 did not show significant differences between control treated types, exception eugenol-treated in which expression MLKL higher controls.
